Function of the AtKUP5 transporter in Arabidopsis thaliana

Potassium is one of the essential elements necessary for plant growth. It is involved in many plant processes, such as osmoregulation, enzymes activaton, etc. These functions are very often closely related to its transport in the cell and the whole plant. Although potassium is abundant in earth's crust, the mount of plant-available form is often insufficient. Potassium deficiency manifests itself on many levels and also contributes to the reduction of yield and quality of agricultural crops. There are many of potassium-transporting proteins in the plant. One of the important families of potassium transporters is the KT/HAK/KUP family. This family includes, among others, the high-affinity transporter HAK5, which is key for the uptake of potassium from the environment with low-potassium availability. One of the not very characterized transporters from the KT/HAK/KUP family is the KUP5 transporter, which I deal with in my diploma thesis. The effect of mycorrhiza on production of plant secondary metabolites

Mycorhizal symbiosis, mutualistic relationship between symbiotic fungus and plant roots, is ranked among important interspecies interactions. It is indicated that about 90% terrestrial vascular plants live in this symbiosis. Arbuscular mycorrhiza (AM) is the most often occurring type of mycorrhiza and simultaneously the most often occurring type of symbiosis of higher plants. Approximately 80% of all of plant species form this symbiosis. By symbiotic coexistence with fungus plant often gains benefits as for example higher tolerance against stress factors, resistance against pathogens or improvement of nutrient uptake. In recent years in series of works is detected that mycorrhiza positively influences production of plant secondary metabolites. This influence can display quantitativelyby increase of metabolite in plant and also qualitatively by change spectrum of substances produced. These substances can have importance in different spheres for human. Medicinal research regularly extends spectum of substances with bioactive effect which have origin just in the plant secondary metabolism. Functioning of arbuscular mycorrhiza in relation to fungal community composition and environmental conditions

Arbuscular mycorrhizal (AM) symbiosis, a widespread plant-fungal relationship, is based on reciprocal resource exchange. The functioning of this fragile relationship balances on the scale from mutualism to parasitism, depending on the specific context. The thesis aims to interlink the functioning of AM symbiosis both with the composition of AM fungal communities and with different abiotic conditions. The thesis is divided into a methodological and a factual part and consists of three publications and one manuscript. All experiments were conducted in greenhouse conditions with medic (Medicago sp.) as host plant. Host plants were inoculated with single AM fungal species in Paper I and II, and with a synthetic AM fungal community of five species in Paper III and IV. The host plant identity, the amount of phosphorus (P) in substrate and the type of substrate played an important role for the achievement of mutualistic AM symbiosis, as demonstrated in Paper I. Paper II showed that mitochondrial and nuclear qPCR markers can be used alternatively for the quantification of particular AM fungal species. However, intraradical fungal biomass was better related to copy numbers of nuclear DNA than of mitochondrial DNA. MICROCLIMATIC INFLUENCES ON WOOD PHENOLOGY OF TREES AND SHRUBS IN TREELINE ECOTONE

This thesis deals with the influence of microclimatic conditions (air temperature near tree/shrub top, temperature of the stem and root zone) on the wood phenology of trees (Picea abies) and shrubs (Pinus mugo) growing on the same site in the alpine tree line (ATL) ecotone. Phenological and microclimatic research was carried out in the Giant Mountains at 1,370 m a. s. l. in the 2017 vegetation period, with the aim to compare the course of wood formation depending on the microclimate, to determine the temperature thresholds of wood growth and differences in the morphology of trees and shrubs. Based on the microclimate analysis of trees and shrubs, individual variables were determined (average, average minimum, average maximum, daily amplitude of air, stem and root zone temperatures) characterizing the growing season. The collection of micro-drills of trees and shrubs took place from April to October in an interval of 6 - 11 days, which were subsequently processed and examined in the laboratory. The following phenophases of wood development were distinguished: cambial, enlarging, wall-thickening, mature cells and cells of the previous ring. Mathematical models of effects of selected factors on photosnythesis for highschools

The goal of this Master thesis was development of teaching materials that are focusing on mastering the fundamentals of mathematical modelling on the examples of photosynthetic processes and influence of selected environmental factors on these processes. Developed models of photosynthetic response to select environmental factors, worksheets, methodical manuals and other supporting materials belong to the teaching materials for secondary schools and serve as a basis of work with models. This teaching materials are aimed to introduce modelling and its significance not only in science but also in education. The present thesis was accomplished on the basis of participation of the Department of Experimental Plant Biology of Faculty of Science of Charles University in Prague in international project NSF within programme GLOBE - The Carbon Cycle. The present thesis is divided into two parts - theoretical part and practical part. The theoretical part includes a literary review and an introduction to the mathematical modelling (in software Stella) that are elaborated considering the teaching purposes and serve as a theoretical background for practical part of present thesis.
